چکیده

Downlo ner and localizer of BRCA2 (PALB2) was originally identified as a BRCA2-interacting protein that is l for key BRCA2 genome caretaker functions. It subsequently became clear that PALB2 was another ni anemia (FA) gene (FANCN), and that monoallelic PALB2 mutations are associated with increased risk st and pancreatic cancer. Mutations in PALB2 have been identified in breast cancer families worldwide, cent studies have shown that PALB2 also interacts with BRCA1. Here, we summarize the molecular and re functions and clinical phenotypes of this key DNA repair pathway component and discuss how its discovery has advanced our knowledge of both FA and adult cancer predisposition. Beyond heredity mutations: alterations in the Fanconi anemia/BRCA pathway and its role in ovarian tumorigenesis

The Fanconi anemia (FA)/BRCA pathway consists of fifteen proteins that mediate DNA homologous recombination (HR) and promote chromosomal stability. Here we review the evidence that genetic and epigenetic alterations in BRCA2 (FANCD1), BRIP1 (FANCJ), FANCD2, FANCF, PALB2 (FANCN) and RAD51C (FANCO), render ovarian cells susceptible to malignant transformation. In addition, we discuss the paradoxi...

Biallelic mutations in BRCA1 cause a new Fanconi anemia subtype.

UNLABELLED Deficiency in BRCA-dependent DNA interstrand crosslink (ICL) repair is intimately connected to breast cancer susceptibility and to the rare developmental syndrome Fanconi anemia. Bona fide Fanconi anemia proteins, BRCA2 (FANCD1), PALB2 (FANCN), and BRIP1 (FANCJ), interact with BRCA1 during ICL repair. However, the lack of detailed phenotypic and cellular characterization of a patient...

The Fanconi anemia/BRCA gene network in zebrafish: embryonic expression and comparative genomics.

Fanconi anemia (FA) is a genetic disease resulting in bone marrow failure, high cancer risks, and infertility, and developmental anomalies including microphthalmia, microcephaly, hypoplastic radius and thumb. Here we present cDNA sequences, genetic mapping, and genomic analyses for the four previously undescribed zebrafish FA genes (fanci, fancj, fancm, and fancn), and show that they reverted t...

No evidence for PALB2 methylation in high-grade serous ovarian cancer

BACKGROUND High-grade serous ovarian cancers are a distinct histological subtype of ovarian cancer often characterised by a dysfunctional BRCA/Fanconi anaemia (BRCA/FA) pathway, which is critical to the homologous recombination DNA repair machinery. An impaired BRCA/FA pathway sensitises tumours to the treatment with DNA cross-linking agents and to PARP inhibitors. The vast majority of inactiva...

Pyrosequencing-based DNA methylation profiling of Fanconi anemia/BRCA pathway genes in laryngeal squamous cell carcinoma.

Fanconi anemia (FA) associated genes [FANCA, -B, -C, FANCD1(BRCA2), -D2, -E, -F, -G, -I, -L, -M, FANCN (PALB2), FANCJ(BRIP1) and FA-linked BRCA1] encode proteins of DNA damage response pathways mutated in FA patients. FA is characterized by congenital malformations, chromosomal instability and high cancer susceptibility. FA patients have a 500-700 times higher risk of head and neck squamous cel...
